Fluorescence immunoassay (FIA) is a simple, rapid, and sensitive technique that is used to quantitevly mesure many compounds including hormones, proteins and drugs. This method has been widely applied in the in vitro diagnostics (IVD) industry.
A modern fluorescent based immunoassay uses as the detection reagent – fluorescent microspores which absorb light or energy (excitation energy) at a specific wavelength and then emits light or energy at a different wavelength.
The FIA test consists of:
1) a conjugate pad containing specific protein (antibody or antigen) conjugated with fluorescent microspheres (probe conjugate) and a control antibody robe conjugate.
2) a nitrocellulose membrane with immobilized proteins in the test area (T line) and a control line (C line).
After adding the specimen to the sample well on the test cassette, protein in the sample will interact with the probe conjugate on the probe pad. The NC membrane diffuses re-solubilized conjugate forward. As the sample moves along the devise binding reagents situated on the NC membrane bind to the target at the T and C line.
Concentration in specimen is positively correlated with the fluorescent density of the fluorescent microspheres, after the measure and calculation of the quantitative fluorescence immunoassay analyzer, then the analyzer would calculate the concentration according to the preset calibration curve in the machine and display the result. C line should appear in any cases, which indicates the reaction system is effective.

Immunoassay is a highly selective bioanalytical method that measures the presence or concentration of analytes including macromolecules and small molecules in a solution through the use of an antibody or an antigen as a biorecognition agent. This method is based on the specificity of antibodies to their antigen. Fluorescent probes, as detectable signals, have been widely used in immunoassay. Fluorescent probes have many advantages as detectable signals including high sensitivity and a variety of measurable properties such as emission intensity, orientation, waveform, and lifetime. In a classical FIA, fluorescent dyes that illuminate in UV light are used to detect the specific binding of an antigen and antibody. The advantages of an FIA method include higher sensitivity detection of the analyte, simplified reagents, and simpler assay designs.
